XNW7201 is an orally available small molecule inhibitor of porcupine (PORCN), a membrane-bound O-acyltransferase essential for the post-translational acylation and secretion of Wnt ligands. By inhibiting PORCN, XNW7201 blocks the Wnt signaling pathway, which is implicated in tumor cell proliferation, survival, and immune evasion. The drug has demonstrated selectivity and favorable pharmacokinetic properties in preclinical studies, with significant inhibitory effects on various gastrointestinal cancers including gastric cancer, colorectal cancer, pancreatic cancer, and esophageal cancer. XNW7201 is being developed primarily for advanced solid tumors and has entered Phase 1 clinical trials to evaluate its safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with locally advanced or metastatic solid tumors[1][2][4][5].
