XNW9015 is a potent, selective, and CNS-penetrating covalent inhibitor of cyclin-dependent kinase 7 (CDK7) developed by Evopoint Biosciences. CDK7 plays a dual role in cell cycle progression as a component of the CDK-activating kinase (CAK) complex and in transcription regulation by phosphorylating RNA polymerase II. In preclinical studies, XNW9015 demonstrated strong antiproliferative activity across various cancer cell lines, including triple-negative breast cancer, ovarian cancer, and colon cancer. It has also shown potential to overcome resistance to CDK4/6 inhibitors in breast cancer models and exhibited superior in vivo antitumor efficacy and pharmacokinetic properties compared to other CDK7 inhibitors like LY3405105.
