XON27 is a first-in-class glyco-humanized polyclonal antibody (GH-pAb) developed by Xenothera for the treatment of hematologic malignancies, including multiple myeloma and B-cell lymphoma. As a polyclonal antibody, XON27 is designed to target multiple antigens simultaneously, which may help overcome resistance mechanisms common in relapsed or refractory blood cancers. Its mechanism of action involves inducing potent cytotoxicity through complement-dependent cytotoxicity (CDC), antibody-dependent cellular phagocytosis (ADCP), and direct induction of apoptosis. Preclinical studies presented at AACR 2024 have demonstrated significant tumor growth reduction in murine models and high tolerability in non-human primates.
