XORLO is a proprietary oral formulation of oxypurinol developed by XORTX Therapeutics. Oxypurinol is a purine-based xanthine oxidase inhibitor (XOI) that reduces uric acid production by inhibiting the enzyme xanthine oxidase. XORLO has been specifically engineered to improve the bioavailability and systemic exposure of oxypurinol compared to non-formulated versions. The drug is being developed for two main indications: - Autosomal Dominant Polycystic Kidney Disease (ADPKD), where it aims to slow kidney function decline by reducing uric acid–mediated injury. - Gout, particularly in patients intolerant to allopurinol. Clinical studies have shown that XORLO achieves higher systemic exposure and improved pharmacokinetics with minimal adverse effects. The product has received orphan drug designation for ADPKD and is advancing through late-stage clinical development for both ADPKD and gout[1][2][5][7].
