XP-8121 is a novel, ready-to-use, once-weekly subcutaneous (SC) injectable formulation of levothyroxine sodium developed using Xeris Biopharma’s proprietary XeriSol non-aqueous formulation technology. It is designed to address the limitations of daily oral levothyroxine therapy for hypothyroidism by providing more consistent thyroid hormone levels and improving patient adherence. Oral administration of levothyroxine can be affected by gastrointestinal absorption variability and food/drug interactions; XP-8121 bypasses these issues with SC delivery. Clinical studies have shown that XP-8121 achieves slower absorption, lower peak plasma concentrations, and extended exposure compared to oral formulations at equivalent doses. The drug acts as an agonist at the thyroid hormone receptor, mimicking endogenous thyroxine (T4) activity to restore normal metabolic function in patients with hypothyroidism[3][4][5][6].
