XR5944 is a novel DNA-targeting small molecule anticancer agent with a unique mechanism of action. It acts as a DNA bis-intercalator, binding preferentially at specific sites (notably 5'-TGCA) within duplex DNA and occupying the major groove. This binding mode disrupts the interaction of transcription factors—most notably the estrogen receptor (ER)—with their consensus DNA sequences, thereby inhibiting ER-mediated transcription. XR5944 was originally developed as a topoisomerase inhibitor but has since been shown to function primarily as a transcription inhibitor by blocking ER-DNA interactions and other major groove-dependent protein-DNA interactions. Preclinical studies have demonstrated potent anti-tumor activity in vitro and in vivo against various human tumor models, including solid tumors[1][2][3][4][5]. The drug was discovered by Professor William A. Denny and coworkers.
