XRN1 inhibitors are a class of small molecule compounds being developed by Accent Therapeutics to target the 5'-3' exoribonuclease 1 (XRN1) enzyme for the treatment of cancer. XRN1 is a key RNA exonuclease responsible for the degradation of single- and double-stranded RNA, playing a critical role in mRNA turnover and the regulation of innate immunity by preventing the accumulation of double-stranded RNA (dsRNA). By inhibiting XRN1, these compounds aim to induce an interferon response in tumors with intrinsically elevated Type I interferon signaling, such as non-small cell lung cancer (NSCLC). These inhibitors are designed to be used as monotherapy or in combination with immune checkpoint inhibitors to enhance anti-tumor immune responses.
