XRN1 shRNA is an experimental RNA-based therapeutic candidate designed to silence the expression of 5'-3' exoribonuclease 1 (XRN1). In pancreatic cancer, XRN1 has been identified as a critical molecular bridge in the KRAS-miR-21 signaling axis, where it facilitates the maturation of the oncogenic microRNA miR-21 at the post-Drosha stage. By depleting XRN1, this shRNA disrupts miR-21 biogenesis, leading to the upregulation of downstream tumor suppressors such as PTEN and PDCD4. Preclinical studies in xenograft and syngeneic models have demonstrated that XRN1 depletion reduces tumor growth and extends survival, highlighting its potential as a targeted strategy for KRAS-driven malignancies.
