XS15 is a synthetic lipopeptide and water-soluble derivative of Pam3Cys, chemically named N-Palmitoyl-S-[2,3-bis(palmitoyloxy)-(2R)-propyl]-Icysteinyl-GDPKHPKSF. It functions as a toll-like receptor 1/2 (TLR1/2) ligand and acts as an immunostimulatory adjuvant in peptide-based therapeutic cancer vaccines. By activating TLR1/2, XS15 induces robust CD8+ and T-helper 1 CD4+ T-cell responses. It has been used to enhance the efficacy of personalized multi-peptide vaccines for hematological malignancies such as acute myeloid leukemia (AML) and chronic lymphocytic leukemia (CLL), as well as in investigational vaccines for solid tumors like fibrolamellar hepatocellular carcinoma. XS15 is typically emulsified with Montanide ISA 51 VG prior to administration[1][4][5].
