XS15-adjuvanted multipeptide vaccine is an experimental therapeutic cancer vaccine developed by the University Hospital Tuebingen for the treatment of glioblastoma multiforme (GBM). The vaccine consists of a cocktail of tumor-associated peptides restricted to HLA-A*02 combined with XS15, a novel, water-soluble lipopeptide that acts as a potent adjuvant by targeting Toll-like receptors 1 and 2 (TLR1/2). This combination is designed to overcome the highly immunosuppressive environment of GBM by inducing robust, long-lasting Th1-polarized CD4+ and CD8+ T-cell responses against tumor-specific antigens. In clinical trials, it is typically administered following standard-of-care radiotherapy and temozolomide in patients with MGMT-methylated tumors.
