XTMAB-16 is a chimeric human-murine monoclonal antibody that specifically targets tumor necrosis factor alpha (TNFα). By blocking TNFα, XTMAB-16 disrupts a key inflammatory pathway implicated in granuloma formation, which is central to the pathogenesis of sarcoidosis. The drug consists of approximately 25% mouse variable region and 75% human constant region, and was developed using a Chinese Hamster Ovary cell line. XTMAB-16 is distinct from other anti-TNF antibodies such as infliximab or adalimumab due to its molecular structure and glycosylation profile, potentially reducing immunogenicity risks. It is being developed primarily for the treatment of pulmonary sarcoidosis with or without extra-pulmonary involvement. Xentria is the developer, and the drug has received Orphan Drug Designation from both FDA and EMA for this indication. Clinical trials are ongoing to assess its safety and efficacy in reducing corticosteroid use among patients with sarcoidosis[1][2][5][6][7].
