Overview

XV-122 is an ultra-long-acting prodrug of dolutegravir, a second-generation HIV integrase strand transfer inhibitor (INSTI). Developed by Exavir Therapeutics using a proprietary prodrug nanomedicine technology, it is designed to extend the dosing interval to four to six months or longer to improve treatment adherence in patients with HIV. The technology involves the chemical modification of the parent drug into a lipophilic prodrug that is then formulated into nanocrystals, allowing for slow release from the injection site. In 2024, ViiV Healthcare acquired the rights to XV-122 through a settlement and license agreement with Exavir, aiming to integrate it into their portfolio of long-acting HIV treatments.
