Overview

XVir-N-31 is a **YB-1-dependent oncolytic adenovirus** (OAV) engineered for tumor-selective replication and destruction of cancer cells, especially glioblastoma. It is designed to replicate predominantly in cells with high nuclear YB-1 expression—a transcription factor upregulated in glioblastoma—and induce oncolytic cell death. Preclinical models demonstrate that XVir-N-31 preferentially targets tumor cells, triggers immunogenic cell death (ICD), and stimulates anti-tumoral immune responses, including increased infiltration of tumor-infiltrating lymphocytes (TILs) and natural killer (NK) cells. The addition of immune checkpoint inhibition (e.g. PD-1/PD-L1 blockade) significantly enhances the therapeutic and abscopal effects of XVir-N-31. The agent has entered phase I clinical trials for patients with recurrent glioblastoma[2].
