Overview

XYA02-8 is a MUC1-C-targeting antibody-drug conjugate (ADC) being developed by Xyone Therapeutics for the treatment of aggressive prostate cancer variants, including castration-resistant prostate cancer (CRPC) and neuroendocrine prostate cancer (NEPC). The drug utilizes the 7B8 monoclonal antibody, which specifically targets the C-terminal subunit of Mucin 1 (MUC1-C), a marker that is significantly upregulated in PSMA-negative and hormone-refractory prostate cancers. Upon binding to MUC1-C, XYA02-8 is efficiently internalized, delivering a cytotoxic payload to the tumor cells. Preclinical studies have demonstrated significant tumor growth inhibition in both androgen-replete and androgen-depleted models, suggesting its potential as a therapeutic option that bypasses the need for systemic hormonal deprivation.
