Drug intelligence / Profile preview
XYG-204 is a water-soluble prodrug of ezogabine (retigabine), a first-in-class Kv7 (KCNQ) potassium channel opener. Developed by Xyzagen, XYG-204 is designed to overcome the pharmacokinetic and formulation limitations of ezogabine, which was withdrawn from the market due to its three-times-daily dosing and risk of tissue discoloration caused by dimer formation. XYG-204 is highly soluble in water and stable in solution, allowing for both oral and parenteral (intramuscular, subcutaneous, and intravenous) administration. In preclinical models, XYG-204 rapidly converts to ezogabine in plasma, providing robust and sustained protection against maximal electroshock-induced seizures. It is being developed for the treatment of focal onset seizures, status epilepticus, and functional neurological seizures.
