XYZ-XI-14 is a novel small molecule sigma-2 receptor ligand developed by researchers at Florida A&M University. It is being investigated for the treatment of solid tumors, specifically triple-negative breast cancer (TNBC), where the sigma-2 receptor is highly overexpressed. In preclinical studies using the TNBC cell line MDA-MB-231, XYZ-XI-14 demonstrated concentration-dependent inhibition of cell viability with an EC50 of approximately 12 µM. At a concentration of 5 µM, the compound inhibits cell proliferation, induces apoptosis, and causes cell cycle arrest at the G0/G1 phase. Additionally, XYZ-XI-14 has been shown to prevent the formation of 3D tumor spheroids and promote the disintegration of preformed spheroids in vitro.
