XZ157 is a potent and selective small molecule inhibitor of the KRAS G12C mutation, developed by Shandong New Era Pharmaceutical Co., Ltd., a subsidiary of Lunan Pharmaceutical Group. KRAS is a member of the RAS family of GTPases that plays a critical role in cell signaling pathways regulating growth and survival. The G12C mutation locks the KRAS protein in an active, GTP-bound state, driving oncogenic signaling in various cancers, particularly non-small cell lung cancer (NSCLC) and colorectal cancer. XZ157 is designed to covalently bind to the cysteine residue at position 12, trapping the protein in its inactive GDP-bound state. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors harboring the KRAS G12C mutation.
