XZP-6877 is an investigational small molecule inhibitor of DNA-dependent protein kinase (DNA-PK) developed by Shandong XuanZhu Pharmaceutical, a subsidiary of Sihuan Pharmaceutical Holdings Group. DNA-PK is a critical enzyme in the non-homologous end joining (NHEJ) pathway, which is the primary mechanism for repairing DNA double-strand breaks in eukaryotic cells. By inhibiting DNA-PK, XZP-6877 blocks the repair of these breaks, thereby sensitizing tumor cells to DNA-damaging agents or inducing synthetic lethality in cancers with specific DNA repair deficiencies. The compound is currently in Phase 1 clinical development for the treatment of advanced solid tumors, including breast cancer.
