Y-27632 is a cell-permeable, highly potent and selective small molecule inhibitor of Rho-associated coiled-coil containing protein kinase (ROCK), specifically targeting both ROCK1 and ROCK2 isoforms by competitively binding to the ATP catalytic site. It is widely used as a biochemical tool in research to study the ROCK signaling pathway, which regulates various cellular functions including smooth muscle contraction, cytoskeletal organization, cell migration, proliferation, and apoptosis. Y-27632 has been shown to inhibit calcium sensitization leading to smooth muscle relaxation, promote survival of human embryonic stem cells and induced pluripotent stem cells during dissociation or cryopreservation, and facilitate differentiation in regenerative medicine applications. It has also been studied for its effects on corneal endothelial cells and cardiac stem cells as well as for use in tissue cryopreservation protocols.
