Y-4

Y-4 is an oral fixed-dose analgesic combination of **pregabalin** and **riluzole** developed by Neurodawn for neuropathic pain. Pregabalin binds voltage-gated calcium-channel alpha-2/delta subunits to reduce release of excitatory neurotransmitters, while riluzole inhibits voltage-gated sodium channels and suppresses excitatory amino-acid-driven neurotransmission. The combination is intended to reduce pain-induced neuronal hyperexcitability through complementary peripheral and central mechanisms. Clinical development includes Phase 2 evaluation in postherpetic neuralgia, alongside completed Chinese Phase 1 studies and a U.S. Phase 1 pharmacokinetic study.
