Overview

This is a combination regimen consisting of **Y101D** (a bispecific antibody targeting PD-L1 and TGF-β), **gemcitabine** (a nucleoside analog antimetabolite), and **nab-paclitaxel** (nanoparticle albumin-bound paclitaxel). Y101D is a humanized bispecific antibody designed to simultaneously inhibit PD-L1 and TGF-β signaling. By blocking PD-L1, Y101D prevents tumor immune evasion by reactivating T cells, while TGF-β inhibition disrupts immunosuppressive tumor stroma and fibrosis, together dual blockade enhances anti-tumor activity and remodels the tumor immune microenvironment[1][3][5]. Gemcitabine is a cytotoxic chemotherapy agent that inhibits DNA synthesis, causing apoptosis in rapidly dividing tumor cells. Nab-paclitaxel is a formulation of paclitaxel bound to albumin nanoparticles, improving delivery to tumors and reducing solvent-related toxicity; it stabilizes microtubules, thereby inhibiting cell division[2][4][6]. As a combination, these agents attack tumors through immunomodulation, disruption of the tumor microenvironment, and direct cytotoxicity. The primary indication is in metastatic or locally advanced solid tumors, with a research focus on metastatic pancreatic cancer and other stroma-rich or immunotherapy-resistant tumors[1][3].
