Y332D is a bispecific antibody designed to simultaneously block the activity of transforming growth factor beta (TGF-β) and vascular endothelial growth factor A (VEGFA). Developed using the Nano-YBODY platform by Wuhan YZY Biopharma, it binds both TGF-β and VEGFA with high affinity, counteracting TGF-β-mediated immunosuppression and VEGF-driven angiogenesis within the tumor microenvironment. Y332D has demonstrated robust antitumor activity in preclinical models, both as monotherapy and synergistically in combination with PD-1 immune checkpoint blockade or radiotherapy, resulting in inhibition of tumor growth, suppression of metastasis, reversal of epithelial-mesenchymal transition (EMT), inhibition of cancer-associated fibroblast activation, and reduction of tumor vessel formation. Its mechanism of action aims to improve the immune milieu and enhance antitumor immune responses in solid tumors[1][2].
