YBL-013 is a preclinical, next‑generation immuno‑oncology bispecific T cell engager developed on Y-Biologics’ ALiCE (Antibody-Like Cell Engager) platform that simultaneously targets programmed death-ligand 1 (PD-L1) on tumor cells and CD3 on T cells to promote localized T cell–mediated killing of solid tumors.[4][5][11][12][13] The molecule uses a unique 2:1 antibody format with bivalent binding to the tumor antigen and a monovalent, lower-affinity CD3-binding arm, designed to concentrate T cell activation at the tumor site, enhance cytotoxicity at low doses, prolong half-life relative to smaller T cell engagers, and reduce systemic and off-tumor toxicity compared with earlier CD3 bispecific designs.[1][4][5][7][12][13] Initially created by Y-Biologics in South Korea and licensed to 3D Medicines (where it is also referred to as 3D057) for development and commercialization in Greater China, YBL-013 is being advanced as a novel therapeutic candidate for a broad range of PD-L1–expressing solid tumors.[1][4][5][8][11][12][14]
