Overview

YCT-529 is an experimental, first-in-class, non-hormonal male contraceptive developed by YourChoice Therapeutics in collaboration with Dr. Gunda Georg at the University of Minnesota. It is an orally administered small molecule that acts as a selective antagonist of the retinoic acid receptor alpha (RAR-alpha), a nuclear receptor involved in vitamin A signaling essential for spermatogenesis. By inhibiting RAR-alpha, YCT-529 disrupts sperm production and release in the testes, leading to temporary infertility without affecting other physiological processes governed by vitamin A. Preclinical studies demonstrated 99% efficacy in preventing pregnancy in mice and significant reduction of sperm counts in non-human primates, with full reversibility after discontinuation and no significant side effects observed. The drug has entered Phase 1 clinical trials to assess safety and efficacy in humans[1][3][4][5][6].
