Overview

Yellow fever virus vaccine is a live-attenuated viral vaccine used to prevent infection by the yellow fever virus, an arbovirus of the genus Flavivirus. The most widely used form is derived from the 17D strain of the virus. It works by stimulating both innate and adaptive immune responses through activation of multiple dendritic cell subsets via Toll-like receptors (TLR2, TLR7, TLR8, and TLR9), leading to production of neutralizing antibodies and cytotoxic T cell responses[6]. Immunity develops in most recipients within ten days and is considered lifelong for nearly all individuals[5]. The primary indication is prevention of yellow fever in people traveling to or living in endemic areas (Africa and South America) or where vaccination is required for entry[1][2][3][8]. The World Health Organization recommends routine immunization in endemic regions. The original 17D strain was developed by Max Theiler in the 1930s through serial passage attenuation[6].
