Drug intelligence / Profile preview
YES1-stabilized CAR-NK is an experimental chimeric antigen receptor natural killer (CAR-NK) cell therapy platform engineered to incorporate YES1 stabilization. Developed by researchers at Tongji University, Shandong University, and Shanghai Pulmonary Hospital, this platform is designed to overcome primary resistance to immunotherapy in non-small cell lung cancer (NSCLC). In the tumor microenvironment, the E3 ubiquitin ligase HUWE1 mediates the ubiquitination-dependent degradation of YES1, a tyrosine-protein kinase critical for NK cell metabolic homeostasis, cytoskeletal organization, and effector function. This degradation is further enhanced in immunosuppressive niches rich in SPP1 (osteopontin). By incorporating YES1 stabilization, these engineered CAR-NK cells resist ubiquitin-mediated degradation, restoring NK cell persistence and cytotoxicity under immunosuppressive conditions.
