YH003

YH003 is a humanized IgG2 agonistic monoclonal antibody that targets CD40, a receptor expressed on antigen-presenting cells. By binding to and activating CD40, YH003 stimulates immune responses and enhances the effector activity of anti-tumor T cells. This immunostimulatory mechanism can transform "cold" tumors (with low immune cell infiltration) into "hot" tumors that are more responsive to immunotherapy. Preclinical studies demonstrated strong anti-tumor effects both as monotherapy and in combination with anti-PD-1 antibodies, without significant hepatotoxicity or other toxicities. Clinically, YH003 has shown good safety and preliminary efficacy in phase I trials for advanced solid tumors when used alone or combined with the PD-1 inhibitor toripalimab. It is currently being evaluated in phase II multi-regional clinical trials for unresectable/metastatic pancreatic ductal adenocarcinoma (PDAC) and melanoma[1][2][3][4][5][7].
