YH012 is a first-in-class bispecific antibody-drug conjugate (BsADC) that targets both Human Epidermal Growth Factor Receptor 2 (HER2) and Trophoblast Cell-Surface Antigen 2 (TROP2). Developed by Biocytogen (and its subsidiary Eucure Biopharma), YH012 consists of a fully human bispecific antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable linker. The drug is designed to treat a variety of solid tumors, including those with low HER2 expression, by leveraging the co-expression of TROP2 to enhance tumor-specific targeting and internalization. Preclinical data presented at AACR 2023 demonstrated superior anti-tumor efficacy compared to parental monoclonal antibodies in models of gastric, colorectal, breast, and non-small cell lung cancer (NSCLC). The dual-targeting approach aims to increase tissue specificity and selectivity while reducing potential toxicity.
