YHC1104 is a preclinical-stage small molecule inhibitor developed by Yuhan Corporation that targets Son of Sevenless homolog 1 (SOS1). SOS1 is a guanine nucleotide exchange factor (GEF) that plays a pivotal role in the activation of RAS proteins (including KRAS, HRAS, and NRAS) by facilitating the exchange of bound GDP for GTP. By inhibiting SOS1, YHC1104 disrupts the activation of the RAS-RAF-MEK-ERK (MAPK) signaling pathway, which is frequently hyperactivated and serves as a primary driver of cell proliferation and survival in various cancers, particularly those harboring KRAS mutations. YHC1104 is currently being investigated as a targeted therapy for the treatment of advanced solid tumors, either as a monotherapy or in potential combination with direct KRAS inhibitors to enhance efficacy and overcome resistance mechanisms.
