Yivoximab (CDX-527) is a recombinant human IgG1 bispecific monoclonal antibody designed to simultaneously target Programmed Death-Ligand 1 (PD-L1) and Cluster of Differentiation 27 (CD27). Developed by Celldex Therapeutics, it utilizes a dual mechanism of action that combines checkpoint inhibition with immune co-stimulation. By blocking the PD-1/PD-L1 inhibitory pathway, yivoximab relieves immune suppression within the tumor microenvironment, while its CD27 agonist activity provides a potent co-stimulatory signal to T cells. This synergistic approach is intended to enhance anti-tumor immune responses more effectively than monotherapy targeting either pathway. The drug is primarily being investigated for the treatment of advanced or metastatic solid tumors that have failed standard therapies.
