YK-BCMA BB-002 is an investigational autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy developed by YaKe Biotechnology for the treatment of relapsed or refractory multiple myeloma. The therapy involves the genetic modification of a patient's own T cells using a lentiviral vector to express a CAR that specifically targets B-cell maturation antigen (BCMA), a protein predominantly expressed on the surface of malignant plasma cells. The CAR construct features a murine-derived anti-BCMA single-chain variable fragment (scFv) for antigen binding, linked to a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. This configuration is designed to trigger T-cell activation, proliferation, and cytotoxic activity upon binding to BCMA, leading to the elimination of myeloma cells. YK-BCMA BB-002 is currently being evaluated in Phase 1 clinical trials.
