YK012 is a bispecific antibody that targets both CD19 and CD3, functioning as a T-cell engager immunotherapy. It is designed to activate T-cell immunity via CD3 while simultaneously targeting CD19, a marker widely expressed during B-cell development. This dual targeting enables the depletion of B cells and aims to reset the B-cell compartment, which is relevant in both autoimmune diseases and certain hematologic malignancies. YK012 has demonstrated milder T cell activation compared to blinatumomab (Blincyto), with extended half-life and limited cytokine release in clinical studies. The drug was initially developed by Excyte Biopharma Ltd (also referred to as Yikester or Yike Si Te) and represents the first bispecific CD19-directed CD3 T cell engager immunotherapy for primary membranous nephropathy (pMN). It is also being investigated for relapsed/refractory non-Hodgkin lymphoma (NHL), acute lymphoblastic leukemia (ALL), and systemic lupus erythematosus (SLE)[1][2][5][6][8].
