Overview

YKL-GB is an E1B 55 kDa-deleted oncolytic adenoviral vector designed as a biologic anticancer agent. It combines the lytic function of an oncolytic adenovirus, which selectively replicates in and destroys tumor cells, with the ability to deliver therapeutic genes. Specifically, YKL-GB expresses both granulocyte-macrophage colony-stimulating factor (GM-CSF) and B7-1 (CD80). GM-CSF is a potent stimulator of antitumor immunity, promoting the maturation of antigen-presenting cells and subsequent T-cell activation. B7-1 is a costimulatory molecule that plays a crucial role in mediating an antitumor response by providing the necessary second signal for T-cell activation, thereby overcoming immune evasion by tumor cells. This dual mechanism aims to enhance the immune response against tumors and provide a potential therapeutic strategy for neoplasia.
