Overview
YKST02 is a first-in-class bispecific antibody developed by Excyte Biopharma for the treatment of multiple myeloma and other B-cell/plasma cell-related diseases. It features a 2+2 symmetric structure based on Excyte’s proprietary FIST (Fusion of IgG and scFv Technology) platform. The drug targets both B-cell maturation antigen (BCMA) and CD3, functioning as a T-cell engager to redirect cytotoxic T cells toward malignant plasma cells expressing BCMA. This mechanism is similar to that of BCMA-targeting CAR-T therapies but delivered as an off-the-shelf biologic. Preclinical data suggest that YKST02 may offer improved safety (notably reduced cytokine release syndrome risk) and efficacy compared to existing agents like teclistamab. The drug is currently in Phase 1 clinical trials for relapsed/refractory multiple myeloma in China, with preclinical development ongoing for IgA nephropathy[1][2][4][5][8].
