YKYY017 is a novel lipopeptide-based, inhaled aerosolized broad-spectrum coronavirus membrane fusion inhibitor. Developed by Yuekang Pharmaceutical (also known as Youcare Pharmaceutical Group) in collaboration with the Institute of Pathogenic Biology at the Chinese Academy of Medical Sciences, the drug targets the heptad repeat 1 (HR1) region of the S2 subunit of the SARS-CoV-2 spike protein. By binding to HR1, YKYY017 prevents the interaction between HR1 and HR2, thereby blocking the formation of the six-helix bundle (6-HB) required for viral membrane fusion with host cells. This mechanism effectively inhibits viral entry into the respiratory tract. YKYY017 is administered via aerosol inhalation and is indicated for both the treatment and prevention of COVID-19. It has completed a Phase 2 clinical trial in China and received IND approval from the U.S. FDA in 2023.
