YL-13027 is a potent, highly selective oral small molecule inhibitor of transforming growth factor-beta receptor 1 (TGFβR1; also known as activin receptor-like kinase 5 or ALK5). It is designed to neutralize TGFβ signaling and enhance immunomodulation in the tumor microenvironment by selectively targeting TGFβR1. By inhibiting this pathway, YL-13027 downregulates TGFβR1-dependent signaling in tumor cells, which can suppress tumor cell proliferation and metastasis. The drug has demonstrated anti-tumor efficacy and immune modulation in preclinical studies and is being evaluated for safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity in clinical trials for advanced solid tumors. It is under development primarily by Yingli Pharma (Shanghai Yingli Pharmaceutical) with collaborations including MD Anderson Cancer Center[1][2][4][5][6].
