YL-18319 is an orally administered, small-molecule poly(ADP-ribose) glycohydrolase (PARG) inhibitor being co-developed by Shanghai Yingli Pharmaceutical and Azkarra Therapeutics. It operates via a synthetic lethality mechanism, selectively targeting and killing tumor cells with specific DNA damage response (DDR) defects, such as BRCA1/2 mutations or homologous recombination deficiency (HRD). Preclinical studies have demonstrated that YL-18319 significantly inhibits tumor growth and induces regression in multiple cancer models, including those resistant to PARP inhibitors. It is currently in Phase 1 clinical development for the treatment of advanced or metastatic solid tumors.
