Overview

YL-23174 is a small molecule inhibitor of Homeodomain-interacting protein kinase 2 (HIPK2) being developed by Yingli Pharmaceutical for the treatment of fibrosis. HIPK2 is a serine/threonine kinase that acts as a central hub in the regulation of fibrotic processes by integrating signals from various pro-fibrotic pathways, including TGF-β, Notch, and Wnt/β-catenin. Overexpression or overactivation of HIPK2 has been linked to the progression of fibrosis in organs such as the kidney and liver. By inhibiting HIPK2, YL-23174 aims to suppress the activation of myofibroblasts and reduce the accumulation of extracellular matrix. The drug is currently in Phase 1 clinical trials in both China and the United States.
