YL205 is a next-generation antibody-drug conjugate (ADC) targeting NaPi2b, a sodium-dependent phosphate transporter (encoded by SLC34A2) that is highly expressed in several cancers, including ovarian, lung, thyroid, and breast cancers. Built on MediLink’s TMALIN platform, YL205 consists of a human monoclonal antibody against NaPi2b conjugated via a protease-cleavable linker to a novel topoisomerase 1 inhibitor payload at a drug-antibody ratio (DAR) of 8. The ADC demonstrates high affinity and specificity for NaPi2b-expressing tumor cells and induces apoptosis upon internalization. Preclinical studies show potent cytotoxicity in vitro and significant anti-tumor activity in vivo with favorable pharmacokinetics and stability. Clinical development is ongoing for advanced solid tumors[4][2][7].
