YM543 (also known as ASP-543) is a potent and selective small molecule inhibitor of sodium-glucose cotransporter 2 (SGLT2), developed for the treatment of type 2 diabetes mellitus. SGLT2 is primarily expressed in the kidney and plays a key role in renal glucose reabsorption. By inhibiting SGLT2, YM543 reduces blood glucose levels by increasing urinary glucose excretion. Preclinical studies demonstrated that oral administration of YM543 significantly reduced hyperglycemia and improved glucose tolerance in diabetic animal models without affecting normoglycemia in healthy animals. The drug advanced to phase II clinical trials for type 2 diabetes mellitus but development was discontinued before approval[4][6][7].
