YMN-A03 is a novel, nebulized bifunctional fusion protein consisting of a nanobody (VHH) targeting programmed cell death 1 ligand 1 (PD-L1) fused to interleukin-15 (IL-15). Developed by West China Hospital of Sichuan University, it is designed for local delivery to the lungs via inhalation to treat advanced non-small cell lung cancer (NSCLC). By targeting PD-L1, YMN-A03 blocks the PD-1/PD-L1 checkpoint pathway, while the IL-15 component promotes the activation and proliferation of T cells and natural killer (NK) cells within the tumor microenvironment. The nebulized formulation aims to achieve high local concentrations in the lung while minimizing systemic toxicity. It is currently being evaluated in Phase I clinical trials for patients with advanced NSCLC who have progressed after standard systemic therapies.
