YN1548 is a novel, orally bioavailable small-molecule agonist of the human glucagon-like peptide-1 receptor (GLP-1R). Developed by Yunovia and Ildong Pharmaceutical, it is intended for the treatment of type 2 diabetes mellitus (T2DM) and obesity. YN1548 functions as a biased agonist, preferentially activating the cyclic adenosine monophosphate (cAMP) signaling pathway over β-arrestin recruitment. In preclinical models, including diabetic monkeys, oral administration of YN1548 significantly reduced blood glucose levels, enhanced insulin secretion, and induced weight loss comparable to the injectable GLP-1 receptor agonist liraglutide. Toxicological evaluations in rats and monkeys have shown the compound to be well-tolerated at high doses, supporting its potential as a convenient oral alternative to peptide-based GLP-1 therapies.
