Overview

YNK105 is an orally delivered small molecule drug containing the R-enantiomer of ketorolac (R-ketorolac), currently in Phase I development by Yinuoke for the treatment of cancer cachexia (cancer anorexia-cachexia syndrome). Unlike the racemic mixture or the S-enantiomer, which is a potent COX inhibitor, R-ketorolac is designed to reduce systemic inflammation and tumor-driven autoimmune responses without the gastrointestinal and renal toxicities typically associated with non-steroidal anti-inflammatory drugs (NSAIDs). Preclinical data suggests YNK105 improves overall survival and maintains body, fat, and muscle weight by reducing plasma interleukin-6 (IL-6) levels and tissue-derived Signal Transducer and Activator of Transcription 3 (STAT3) activity. Yinuoke approaches cachexia as a tumor-driven autoimmune disorder rather than a purely metabolic condition.
