YNW-1 is a dual-warhead, bident small molecule inhibitor designed to target the Epidermal Growth Factor Receptor (EGFR). It is specifically engineered to overcome resistance in non-small cell lung cancer (NSCLC) caused by mutations at the C797 site, which typically render third-generation inhibitors like osimertinib ineffective. YNW-1 achieves this by simultaneously and covalently binding to two distinct residues within the EGFR ATP-binding pocket: cysteine 775 (C775) and cysteine 797 (C797). This dual-targeting strategy ensures that a single-site mutation at either cysteine is insufficient to confer resistance, maintaining the drug's inhibitory potency. Developed by researchers at institutions including the Dana-Farber Cancer Institute, YNW-1 represents a next-generation approach to EGFR-targeted therapy.
