yoda1

Yoda1 is a synthetic small molecule and the first known agonist of the mechanosensitive ion channel Piezo1. Piezo1 is involved in various physiological processes including regulation of blood pressure and red blood cell volume. Yoda1 activates purified Piezo1 channels in both human and mouse systems by eliciting calcium flux specifically in cells expressing Piezo1. It has been widely used as a research tool to study mechanotransduction pathways and the role of Piezo1 in vascular biology, neurobiology, lymphatic function, and disease models such as Alzheimer's disease. In preclinical studies, Yoda1 has demonstrated effects on endothelial signaling (e.g., Akt/ERK pathways), inhibition of Rac1-mediated actin rearrangement/macropinocytosis, improvement of brain lymphatic drainage functions related to neurodegenerative diseases like Alzheimer's disease, and modulation of cell survival/proliferation signaling[2][3][6].
