Youdujing (YDJ) is a traditional Chinese herbal formula developed as an in-hospital preparation, primarily utilized for the treatment of diseases associated with human papillomavirus (HPV) infection. It has been clinically used for over 20 years to treat conditions such as cervical precancerous lesions, recurrent condyloma acuminata, and flat warts. The preparation is typically formulated as an external lotion or cream for topical and vaginal application. Mechanistically, YDJ extract induces necroptosis in cervical cancer cells by activating the receptor-interacting protein kinase 1 (RIP1) pathway, promoting the formation of the necrosome complex (RIP1/RIP3/MLKL). Furthermore, it has been shown to reverse cervical precancerous changes and clear high-risk HPV infections by down-regulating the mRNA expression of human telomerase reverse transcriptase (hTERT), thereby reducing telomerase activity.
