YP218 Fv-PE38 is a recombinant fusion immunotoxin consisting of the variable fragment (Fv) of the rabbit monoclonal antibody YP218, which targets a rare epitope in the membrane-proximal region (region III, amino acids 525–560) of human mesothelin, fused to the PE38 fragment of Pseudomonas exotoxin A. This construct is designed to selectively bind to mesothelin-expressing tumor cells (notably in mesothelioma and other solid tumors) and induce cell death via toxin-mediated inhibition of protein synthesis. YP218 Fv-PE38 has demonstrated potent anti-tumor cytotoxicity in vitro against primary mesothelioma cell lines and significant anti-tumor effects in vivo in a xenograft mouse model. The parent YP218 antibody has also served as the basis for several antibody therapeutics and engineered cell therapies[2][3][1][10].
