Drug intelligence / Profile preview
YR-D76 is a preclinical small molecule dihydroorotate dehydrogenase (DHODH) inhibitor being developed by Sichuan Enray Pharmaceutical Sciences. It targets the coenzyme Q (CoQ) binding domain of the mitochondrial DHODH enzyme. By interfering with the ubiquinone (CoQ) reduction process, YR-D76 disrupts the mitochondrial ferroptosis defense pathway, inducing a lipid peroxidation storm that leads to ferroptotic cell death. Preclinical studies have demonstrated that YR-D76 has significant antitumor efficacy against refractory solid tumors, including pancreatic cancer.
