Overview

YR-G346 is a preclinical small molecule glutathione peroxidase 4 (GPX4) inhibitor being developed by Sichuan Enray Pharmaceutical Sciences. It is designed to target the active site of selenocysteine in the GPX4 protein, thereby blocking the glutathione regeneration pathway and promoting ferroptosis (iron-dependent cell death) in tumor cells. Preclinical studies have demonstrated that YR-G346 possesses superior metabolic stability and tumor penetration compared to competing candidates in its class. The program is currently in preclinical development, with IND-enabling studies planned by 2028 for the treatment of oncology indications.
