Overview

YS-01 is a novel, potent, and selective small-molecule inhibitor of pendrin (SLC26A4), an anion exchanger that mediates the transport of chloride, bicarbonate, iodide, and other anions. Identified through high-throughput screening of synthetic compounds, YS-01 has demonstrated significant therapeutic potential in preclinical models of inflammatory airway diseases and acute lung injury. It potently inhibits pendrin-mediated anion exchange activity (such as Cl-/SCN- and Cl-/I- exchange) and reduces pendrin protein expression. In animal models, YS-01 attenuates airway hyperresponsiveness, goblet cell hyperplasia, and mucin expression in asthma models, and alleviates lipopolysaccharide (LPS)-induced and ventilator-induced lung injury (VILI) by suppressing pro-inflammatory cytokine production and NF-κB activation.
